Human MDR1 gene encoding P-glycoprotein (P-gp) plays an important role in multidrug resistance to chemotherapeutic agents as well as affects the pharmacokinetics of many drugs and xenobiotics. To date, over 60 single nucleotide polymorphisms (SNPs) have been reported for the coding region of MDR1 gene, some of which can affect P-gp expression and function. However, the effect of these polymorphisms on P-gp function and their clinical impact are in most cases unclear. In this review, three most common MDR1 SNPs (C1236T, G2677T/A and C3435T) and their potential implications for cancer pharmacogenetics are summarized. © 2010 by Türkiye Klinikleri.
