Effect of different diluents on the motility of turkey semen during cryopreservation

Özet

The aim of this study was to investigate the effect of different extenders on sperm motility in cryopreservation of turkey semen. Semen was collected from five toms by manual massage method twice a week for five weeks. Some spermatological parameters of collected semen was assessed and mean ejaculate volume (ml), sperm motility (%) and sperm concentration (x 10(9)/ml) were recorded as 0.28 +/- 2.8, 84.8 +/- 1.12 and 5.1 +/- 0.34 respectively. Pooled semen were divided into four equal parts and diluted 1:3 with extenders (Graham, Glucose, Ringer and Tris + Glucose) containing 10% Dimethyl sulphoxide (DMSO) and equilibrated at 4 T for 90 min. Samples were frozen at -120 degrees C on liquid nitrogen vapour for 15 min. Sperm motility after dilution was averaged 74 +/- 2.6, 78 +/- 2.0, 84 +/- 2.2, 62 +/- 2.9 % and post-thaw motility was 31 +/- 2.4, 40.5 +/- 2.8, 17 +/- 2.0, 11 +/- 1.4 % respectively. According to the obtained results, glucose extender provided better environment for turkey semen undergoing cryopreservation than the others (p < 0.001).
