Effect of different genotypes and auxins on efficient somatic embryogenesis from immature zygotic embryo explants of maize

Özet

Efficient somatic embryogenesis has been established from immature embryos of three different maize cultivars and an ecotype cultured on nutrient media supplemented with various concentrations of 2,4-dichlorophenoxy acetic acid (2,4-D), picloram and dicamba. Embryogenesis frequency was influenced by the genotype and auxins tested. Considering both the percentage of explants producing somatic embryos and the number of embryos per explant, lower levels of 2,4-D (0.5 or 1 mg/l) induced the highest frequency of somatic embryogenesis in cultivars "3394 " and " 7993"; whereas embryo formation in ecotype "kirmizi cin" and cultivar "3377" was best achieved on media containing higher concentrations (4 or 8 mg/l) of picloram. Somatic embryos were developed into plantlets on regeneration medium and individual plantlets were then transplanted to compost in small pots. They were first acclimatized to ambient conditions in high relative humidity in growth cabinet and later established in large open pots under greenhouse conditions.
