Spectrofluorimetric Determination of Telmisartan and Amlodipine by Multıvariate Curve Resolution

Özet

Hypertensive patients usually need to use two drug substances to keep blood pressures under control. Pharmaceutical preparations containing telmisartan and amlodipine are used for this purpose [1]. Accurate, precise and economical analytical methods are in need to simultaneously quantify drugs in pharmaceutical preparations. Although telmisartan and amlodipine both have fluorescence properties, classical spectrofluorometric analysis of these drugs in their mixtures was not possible due to overlapping excitation and emission spectral bands in same wavelength region. In order to overcome this problem, multivariate curve resolution was applied to the excitation-emission dataset for the determination of telmisartan and amlodipine in tablets without any chemical pretreatment or separation process.

Excitation-emission matrices of the samples containing both drugs in the concentration range of 0.25 - 4.50 μg/mL were recorded and used to obtain a three-way data array. Decomposition of this data array was performed by multivariate curve resolution to obtain the estimated values of telmisartan and amlodipine content. Figure 1 shows the estimated intensity of telmisartan (TS) and amlodipine (AD) in all samples.

Calibration and quantification steps were done by using the estimated intensities. Validation of the method was performed by analyzing synthetic mixtures, standard addition samples, inter-day and intra-day samples. The proposed method was then applied to the analysis of tablet samples. The assay results and their relative standard deviations were reported as 39.59 and 2.39 for telmisartan and 4.93 and 2.02 for amlodipine. (Label claim: 40 mg telmisartan and 10 mg amlodipine per tablet). The successful results showed that the proposed method is suitable for the fast and direct simultaneous analysis of telmisartan and amlodipine in tablets.
