Özet

Recently a new group of cardiotonic drugs called phosphodiesterase inhibitors is being widely used in medical practice. The present study was designed to examine the effects of phosphodiesterase inhibitors on the negative inotropy induced by thiopenthone. Fifteen frogs, weighted 75-100 g, were used in this study. The stripes obtained from the ventricules were placed into tissue bath containing Krebs solution. One end of the muscle was fixed at the organ hanger while the other end was connected to force- displacement transducer and then negative inotropy was evoked by adding thiopenthone and carbachol in tissue bath. Theophylline, amrinon, sulmasole and zaprinast of which phosphodiesterase inhibitors were added into the tissue bath in cumulative doses and the effects of phosphodiesterase inhibitors on negative inotropy were recorded. As a result we concluded that theophylline, amrinone and zaprinast had no effect on reversing negative inotropy induced by thiopenthone and carbachol whereas sulmasole restored negative inotropy and increased ventricular contractions.
