Aim: This study aims to investigate the possible involvement of nitric oxide synthase (NOS) in cisplatin-induced oxidant stress and nephrotoxicity. Methods: Blood samples and kidneys of 7 animals for the control group and 14 animals treated with a single dose of cisplatin (5 mg/kg body weight) for the experimental group were used throughout the study. NOS activity and malondialdehyde (MDA) levels were measured in the renal tissue. Results: NOS activity was found to be suppressed in renal tissue after 24 h of cisplatin treatment but, to be increased after one week of treatment. Renal MDA level was however high after 24 h and low after one week of treatment. Interestingly, NOS and MDA values were in contrast in the renal tissues. Conclusion: Results suggest that cisplatin at the beginning suppresses and then, induces NOS activity in renal tissue. These changes in renal NOS activity may be involved in the oxidant stress created by cisplatin in the renal tissue and in the cisplatin-induced nephrotoxicity.
