Bioavailability and pharmacokinetics of oxytetracycline in cultured rainbow trout following intramuscular administration

Özet

The intramuscular bioavailability of oxytetracycline (OTC) was investigated in rainbow trout in freshwater at 10.5°C. A group of 70 fish received OTC, an intravenous injection of 5 mg/kg body weight. To the second group (n=105) OTC was administered intramuscularly at a dose rate of 50 mg/kg body weight. OTC plasma concentrations were determined by high performance liquid chromatography (HPLC). The pharmacokinetic parameters were described by a two-compartment open model giving distribution half-lives of 0.907 hand 0.751 h, and elimination half-lives of 33.5 h and 24.264 h for i.v. and i.m. administration of OTC, respectively. After i.v. administration of OTC, other pharmacokinetic parameters estimated were area under the curve (AVC : 145.078±4.45 μg h/ml) and mean residence time (MRT : 40.604±1.150 h). Following i.m. administration, AUC was 1116.184±98.332 μg h/ml, MRT was 34.55±0.364 h, Cmax was 41.542 μg/ml and T max was 2 h. The i.m. bioavailability (F) of OTC in rainbow trout was found to be only 76.93%.
