Molecular Characterization of Apple mosaic virus in Turkish and Ukrainian Isolates

Özet

One of the most important factors that limit the production of apple and hazelnut in Turkey is Apple mosaic ilarvirus (ApMV). The patogen is easily distributed with vegetative plant materials and it is gradually spreading in both pome fruits, hazelnut and oil rose plantations in Turkey Apple mosaic virus isolates were collected from mainly apple and hazelnut grown areas and presence of the pathogen was detected by DAS-ELISA and RT-PCR amplification. Surveys were conducted to apple plantations of Ukraine during the years 2008-2009 and ApMV infected samples were collected. These isolates were compared with the isolates maintained at Ankara University, Faculty of Agriculture, Department of Plant Protection. Silica method was applied for total RNA isolation and 262 bp long PCR products were obtained in RT-PCR tests. All of the isolates were subjected to enzymatic cleavage by Hinf I, BamH I, Ava I, EcoR I, Ssp I, Rsa I, Hind III, Hinc II restriction enzymes and only with Ava I, BamH I, Hinf I, Ssp I enzymes cuttings were accomplished and phylogenetic analysis was performed. Phylogenetic analysis result showed that Turkish isolates of ApMV were composed of 9 and Ukrainian apple isolates were composed of 6 genotypes. When sequence data of apple isolates of both countries were compared, homology in sequences were detected.
