Abstract

The aim of this study was to investigate whether the deficiency uridine monophosphate syntase (DUMPS) allele exists in Holsteins cattle breed including native cattle breeds in Turkey. DUMPS is an autosomal recessive hereditary disease resulting embryonic death. Animal material of this study consisted of 120 Turkish Holstein, 20 Swiss Brown, 20 Native Black, 20 Gray Steppe, 20 South Anatolian Red, 20 East Anatolian Red breeds of cattle. Turkish Holsteins were breeding bulls and young progeny test bulls in different providences of Turkey. DNA materials were isolated from blood samples using phenol chloroform extraction method. The extracted DNA was amplified by PCR and than digested by Ava I endonuclease for the detection of the mutation sites. The digested PCR products were subjected to electrophoresis using 4% NuSieve agarose gel. There were no positive results indicating the presence of DUMPS allele in any sample from the breeds and samples studied.
