The protein phosphorylation patterns in human oocytes have been analyzed using MPM2 antibody to characterize the nucleo-cytoplasmic modifications during early meiotic stages. Results show that, in GV stage oocytes, protein phosphorylation is restricted to a few foci in nucleoplasm whereas by the onset of prometaphase a sudden burst of cytoplasmic phosphorylation occurs. There is a gradual increase in cytoplasmic phosphorylation during further meiotic stages that seems to be correlated with increased MPF activity. Unlike in other species, no evidence for cytoplasmic phosphorylated foci or chromatin/chromosome labeling was noted in all samples of human oocytes examined.
