Qualitative analysis of phenolic compounds in olive cuttings

Özet

Cuttings of cvs. Gemlik and Domat were placed in the Shaded Polyethylene Tunnel (SPT) following various treatments for rooting. Samples for each cultivar and treatment were collected from SPT, at 15-day intervals and prepared for analysis by separating the wood, cortex and leaves of each cutting. Qualitative analysis of phenolic compounds in each part of the cuttings was done using a thin layer chromatography (TLC) method. TLC plates were examined by a video documentation system and results were quantitated using bitmap-densitometric graphic analysis system. A greater condensation of chlorogenic acid (CA) was present in all parts of cuttings of cv. Domat than in those of cv. Gemlik. All treatments of both cultivars showed a strong reduction in CA condensation in the second period of sampling (the 30th day) in 1995, CA condensation generally occurred in samples subjected to wounding and to wounding+IBA treatments, with increment on the 45th 60th and 75th day of sampling during the rooting study.
