Özet

Short-term semen storage is a valuable tool for the distribution of semen or cryopreservation of gamete cells for AI programmes. To implement reproductive technologies for the conservation of wild deer population in Turkey, we aimed to determine the longevity of short-term stored follow deer sperm with two different diluents. We used five males aged between 2 and 4 years kept in Ankara Sincan Zoo in this study. Semen was collected by using an electro- ejaculator in breeding season (November) and samples were divided into two aliquots, diluted with either skimmed milk (SM) or Tris up to 10 × 106 spermatozoa/mL, and stored at +4°C for 12 days. The motil- ity of the samples was recorded on Day 1, 2, 3, 4, 5, 7, 10, 11 and 12 by using computer-assisted sperm analyser (CASA). The statis- tical analysis was performed using the MIXED procedure in SPSS, and descriptive statistics including the mean and standard deviation were calculated to summarize the data. The mean values of 12 days motility with Tris and SM extenders were found 36.65 ± 11.0 and 46.87 ± 11.1 (p = 0.007) respectively. Besides, the average longev- ity was significantly similar between the first day (61.57 ± 12.0) and 2–10 days (54.95 ± 12.0 to 39.69 ± 12.0) while a significant decrease was observed on day 12 (12.34 ± 2.0) (p < 0.001). The motility of deer sperm can be maintained up to 11 days during short-term storage. Although both diluents can be used for deer sperm, SM is suggested due to its higher protection of the motility.
