Hayvan yetiştiriciliği ile yayılan metan gazı, sera gazı emisyonuna katkıda bulunmaktadır. Rasyonların
kaba ve konsantre yem oranları ile kullanılan yem katkı maddelerinin metan üretimine etkisinin ortaya konması,
sera gazını azaltmaya yönelik stratejileri belirlemede yardımcı olabilmektedir. Bu çalışmada, farklı kaba ve
konsantre yem oranlarına ilave edilen maya hücre duvarının in vitro ruminal gaz ve metan üretimine etkisini
belirlemek amaçlanmıştır. Bunun için 2 adet kanüllü Holstein ırkı inekten alınan rumen sıvıları hemen
laboratuvara getirilmiştir ve tampon solüsyonu ile karıştırılarak HFT sisteminde 48 saat boyunca inkübasyona
bırakılmıştır. Denemede 3 x 2 faktöriyel tasarım kullanılmış olup 3 farklı kaba konsantre yem oranı (30:70;
50:50; 70:30) oluşturularak maya hücre duvarı (+, -) ilave edilmiştir. İnkübasyonun 2., 6., 12., 24. ve 48.
saatlerinde üretilen gaz miktarları kayıt altına alınmıştır. İnkübasyon sonunda (48 h) alınan gaz örneklerinde
metan gazı miktarı tespit edilmiştir. Kaba:konsantre yem oranının gaz üretimi üzerine etkisine bakıldığında;
30:70 oranlı gruplarda 6. ve 12. saatlerde gaz üretiminde artış gözlemlenirken (P<0.05), 70:30 oranlı gruplarda
metan gazı miktarı üzerine azaltıcı etkisi olduğu (P<0.05) görülmüştür. Maya hücre duvarının gaz üretimi
üzerine etkisine bakıldığında; 6., 24. ve 48. saatlerde gaz üretiminde artış gözlemlenirken (P<0.05) metan gazı
üzerinde azaltıcı etkisi olduğu (P<0.05) görülmüştür. Yem ve maya hücre duvarının etkileşimi incelendiğinde
2., 6., 12. ve 24. saatlerde gaz üretiminde artış gözlemlenirken (P<0.05) metan gazı üzerine bir etkisi olmadığı
(P>0.05) görülmüştür. Sonuç olarak maya hücre duvarının farklı kaba konsantre yem oranlarında metan gazını
azaltıcı yönde etki yaptığı, özellikle 70:30 oranındaki yemlere ilave edilmesinin en etkili olduğu görülmüştür.

Methane gas emitted by animal husbandry contributes to greenhouse gas emissions. Revealing the the
effect of feed additives with forage:concentrate feed ratios of the diets on methane production can help
determine strategies to reduce greenhouse gasses. In this study, it was aimed to determine the effect of yeast cell
wall on in vitro ruminal gas and methane production at different roughage:concentrate ratios. For this purpose,
rumen fluids were collected from 2 cannulated Holstein cows, immediately transferred to the laboratory and
mixed with buffer solution and incubated in the HFT system for 48 hours. In the trial, 3 x 2 factorial design was
used and yeast cell wall (+, -) added by creating 3 different roughage:concentrate ratios (30:70; 50:50; 70:30).
The gas produced at the 2, 6, 12, 24 and 48h of the incubation was recorded. The methane gas production was
determined in the gas samples at 48h of incubation. The increasing effect of forage: concentrate ratio (30:70) on
gas production was observed at 6 and 12h of incubation (P<0.05),while methane production reduced in the
70:30 ratio groups (P<0.05). While yeast cell wall increased the gas production at the 24h and 48h (P<0.05), it
decreased the methane gas production (P<0.05). When the interaction of feed and yeast cell wall was examined,
it was observed that gas production increased (P<0.05) at the 2, 6, 12 and 24h of incubation, while it did not
affect the methane gas production (P>0.05). As a result, it was observed that the yeast cell wall had a reducing
effect on methane gas at different forage to concentrate ratios, and it was most effective when supplemented to
the diets at a ratio of 70:30.
