Simultaneous determination of phenylephrine hydrochlortoe and chlorpheniramine maleate by ratio spectra derivative spectrophotometry and high - performance liquid chromatography

Özet

Two methods are described for the simultaneous determination of phenylephrine HCl (I) and chlorpheniramine maleate (II) in combined pharmaceutical dosage forms. Using the first method , ratio spectra derivative spectrophotometry,the signals were measured at 238.9 nm and 244.4 nm for chlorpheniramine maleate and at 280.0 nm and 286.4 nm for phenylephrine HCl in the mixture , in the first derivative of the ratio spectra. In this method, ratio spectra derivative spectrophotometry, the calibration graphs for 30.0 - 60.0 *g.ml-1 phenylephrine HCl and for 24.0 - 54.0 *g.ml-1 chlorpheniramine maleate were obtained. The second method is based on high-performance liquid chromatography on LiChrosorb RP- C18 column(5 micron, 20 cm×4.6 mm) using IM orthophosphoric acid:methanol:water (34:5:61 v/v ) with a mobile phase at a flow rate of 1.2 ml/min.Detection was carried out using a UV detector at 269.0 nm. Both methods showed good linearity, precision and reproducibility. The proposed methods were successfully applied to the determination of these drugs in laboratory-prepared mixtures and in tablets.
