A high frequency bulblet regeneration protocol was developed for ornamental plant Ornithogalum platyphyllum using immature embryos. Immature embryos of O. platyphyllum were cultured on Murashige and Skoog medium (MS) supplemented with different concentrations of N-6-benzylamino-purine (BAP), Kinetin (KIN) and alpha-naphthaleneacetic acid (NAA) with or without pretreatment at 4 degrees C for a week. Large numbers of bulblets (over 14 bulblets/explants) were obtained front without pre-treated immature embryos on a MS medium supplemented with 1 mg l(-1) BAP after 10 months of culture initiation. Pre-treatment of immature embryos at 4 degrees C reduced the bulblet regeneration. Rooted bulblets were transplanted to pots containing potting mixture and grown in a growth chamber and total 175 regenerated plants were acclimatized. High survival ratio (100%) was achieved.
