Özet

This study was aimed to determine the effects of sodium hypochlorite solutions at different concentrations (2.5, 3.75 and 5.0%), temperatures (25, 35 and 45 degrees C) and application periods (10, 15 and 20 min) on seed establishment, dormancy-breaking and seedling growth of Lathyrus chrysanthus seeds to obtain healthy uninfected seedlings in large quantities under in vitro conditions. First, the optimum concentration and application period of NaOCl were determined as 3.75% and 15 min at a constant temperature of 25 degrees C. Then, the optimum temperature of NaOCl was tried to be determined. According to the results, all parameters were negatively affected by higher concentration, temperature and application period of NaOCl over 3.75%, 35 degrees C and 15 min, respectively. The highest value with respect to seed germination and obtained seedling percentages, seedling and root lengths, seedling fresh and dry weights, chlorophyll a, chlorophyll b and total chlorophyll contents, number of stomata and cell in the field of view area, stomata width and length, and cell width were obtained when NaOCl was used at 3.75% concentration, 35 degrees C temperature and 15 min application period.
