Özet

The study reports in vitro regeneration of an important forage and hay plant Trifolium pannonicum JACQ. subsp. elongation (WILLD.) which is endemic to Turkey and is popularly used by the Anatolian people as traditional forage. The study was carried out primarily to aid in the development of and efficient micro-propagation method to assist breeding using hypocotyl cotyledon node and cotyledon base as explants. The highest frequency of shoot regeneration (100%) and number of shoots per explant (20.33) with shoot length of 3.50 cm were recorded from hypocotyl explants on MS medium containing 1 mg/l BAP-0.4 mg/l anapthaleneacetic acid (NAA). Rooting was achieved by culturing the shoots on MS medium containing different of indole-3-butyric acid (IBA). Maximum frequency of rooting was recorded on MS medium containing 0.5 or 0.75 mg/l BAP-0.4 These plants were transferred to pots and acclimatized in the greenhouse, where they flowered and set seed.
