In vivo and in vitro evaluation of Saanen buck semen frozen with different antioxidants

Özet

In this study, it was aimed to investigate the effect off principle sperm characteristics and fertility parameters of freezing of Saanen buck semen with skimmed milk powder extender containing different antioxidants. Ejaculates collected by artificial vagina from a total of 4 Saanen buck were used in the study. Principle spermatological characteristcs were determined in semen collected as double ejaculation and ejaculates having normospermie quality were pooled. Semen samples were evaluated as split ejeculate in the experiment and control groups. Split samples were extented with skimmed milk powder (SMP) extender containing 5% glycerol, 10% egg yolk and supplemented with different antioxidants (5 mM cysteine and 1000 mu g/ml hyaluronic acid). Straws contain extended semen was equilibrated at +4 degrees C for 2 h, frozen in vapor of (15 min at -120 degrees C) liquid nitrogen and stored in liquid nitrogen. Frozen semen was thawed in a water bath at 37 degrees C for 30 seconds. According to post-thawing assessments, the sperm motilities in semen extended with skimmed milk powder containing antioxidants cysteine or hyaluronic acid or Without any antioxidant (control group) were recorded as 44.0%, 30.5% and 39.5% respectively. The percentage of abnormal spermatozoa and abnormal acrosome were found 52.8%, 53.6%, 59.1%, 35.9%, 38.3%, 40.4% respectively. The percentage of dead spermatozoa was recorded as 48.8%, 58.1%, 53.8%. Regarding the spermatozoal motility, it was significantly (p < 0.01) higher for the extenders containing cysteine or skimmed-milk powder (control) as compared to the extender containing hyaluronic acid, however there was no Such difference (p > 0.05) between the there extenders given based off the rates of dead spermatozoa, abnormal spermatozoa and abnormal acrosome. Pregnancy and parturition rates obtained from semen frozen with skimmed milk powder extender contains cysteine, hyaluronic acid and no additive (control) were determined 50%, 50%, 40% and 33.3%, 16.6%. 20% respectively. No significant differences were determined statistically between pregnancy and parturition rates in goat which were inseminated with cysteine, hyaluronic acid and skimmed milk powder (control) extenders (p > 0.05). In conclusion, it was determined that cysteine had a favourable effect upon the improvement of spermatological characteristics and fertility parameters.
