Adventitious shoot regeneration in sainfoin (Onobrychis viciifolia Scop.)

Abstract

Cotyledon and hypocotyl explants from in vitro grown seedlings of sainfoin were cultured on a range of media to develop an adventitious shoot regeneration system suitable for transformation. In general, callus formation was not influenced by the explants used, however, hypocotyls appeared to have a better regeneration capacity than cotyledons. The highest frequency of shoot formation was achieved from hypocotyl segments cultured onto the Murashige and Skoog (MS) medium containing 0.5 mg/l 6-benzylaminopurine (BAP) and 0.2 mg/l α-naphthaleneacetic acid (NAA). Regenerated shoots rooted in half strength MS supplemented with either 1 mg/l indole-butyric acid (IBA) or 1 mg/l NAA.
