In this study, the simultaneous determination of chlordiazepoxide (I) with clidinium bromide (II) and amitriptyline hydrochloride (III) in its binary mixtures was realized using ratio spectra derivative spectrophotometry. The signals were measured at 256.7 and 221.8 nm in the I / II mixture and at 275.1 and 218.0 nm in the I / III mixture for I and its co-existing compounds, respectively, in the first derivative of the ratio spectra of the solutions of these mixtures in methanol. The concentration ranges for Beer's law compliance and the relative standard deviations of the method were found to be, in turn, 6.0 - 20.0 μgml-1, 0.32 % for I and 8.0 - 20.0 μgml-1, 0.93 % for II in the first mixture and 6.0 - 20.0 μgml-1, 0.32 % for I and 4.0 - 20.0 μgml-1, 1.23 % for III in the second mixture. The method was successfully applied to the tablets containing the above-mentioned drug combinations without any interference by the excipients.
