Özet:Bu çalışma, bir ACE inhibitörü olan perindopril'in kalp yetmezlikli ve hipertansiyonlu köpeklerde etkinliğini araştırmak amacıyla, Ankara Üniversitesi Veteriner Fakültesi İç Hastalıkları Anabilim Dalı kliniğine getirilen, değişik kilo ve yaşlarda, farklı ırk ve cinsiyette kalp yetmezlikli ve hipertansiyonlu 20 köpek (deneme grubu), farklı ırk ve kiloda her iki cinsiyetten sağlıklı 10 köpek (kontrol grubu) olmak üzere toplam 30 köpekte yapıldı.Çalışmada kan basınç ölçümleri deneme grubundaki köpeklerde 0.,1.,7., 14., 21. ve 28. günler olmak üzere altı kez, kontrol grubunda ise 0. ve 28. günler olmak üzere iki kez yapıldı. Köpeklerin kan basınçları (SYS, DİA ve MAP değerleri) veteriner alanda kullanım sertifikalı köpeklere spesifik memodiagnostic tansiyon cihazıyla osilometrik yöntemle ölçüldü. Deneme grubunu oluşturan köpeklerde deneme süresince antihipertansif olarak günde tek sefer oral olarak 0,5 mg/kg dozda perindopril ve üç gün süreyle 1 mg/kg dozda i.m. furosemid kullanıldı. Deneme ve kontrol grubunu oluşturan köpeklerin 0. ve 28. günlerde olmak üzere iki kez hematolojik ve serum biyokimyasal ölçümleri yapıldı. Hemogram için alınan örneklerden; Hgb miktarı, RBC, PLT ve WBC sayıları, serum biyokimyası kontrolleri için CK, LDH, AST, CK-MB ve Kardiak Troponin-I enzim ve Na+, K+, Cl- iyon değerleri belirlendi.Kontrol grubunu oluşturan köpeklerin fiziksel muayenelerinde kalp yetmezliği ve hipertansiyonun varlığını gösteren herhangi bir bulguya rastlanmadı.Çalışmanın ilk günü kalp yetmezliği ve hipertansiyon belirlenen deneme grubu tüm köpeklerin SYS, DİA ve MAP değerlerinin, kontrol grubunu oluşturan köpeklerin kan basıncı değerlerinden oldukça yüksek olduğu belirlendi. İlaç uygulamasının 28. gününde ise; deneme grubunu oluşturan bütün köpeklerin SYS, DİA, MAP değerlerinin, kontrol grubunu oluşturan köpeklerin kan basıncı değerlerine yakın olduğu ve fizyolojik sınırlarda olduğu belirlendi.Köpeklerde perindopril kullanımının kan tablosu ve kan serumu biyokimyasal değerlerinde önemli değişiklikler oluşturmadığı belirlendi.Bu çalışma ile kalp yetmezliği ve hipertansiyon belirlenen köpeklerde hipertansiyonun tedavisinde bir ACE inhibitörü olan perindopril'in etkili olduğu, kan basıncının normal seviyede sürdürülmesi için hergün oral olarak 0,5 mg/kg dozda kullanımının uygun olduğu sonucuna varıldı. AbstractThis study is to investiage the effectiveness of perindopril, an ACE inhibitor, on hypertensive and heart failure in dogs brought to Ankara University Faculty of Veterinary Medicine Department of Internal Medicine clinic. There were total of 30 dogs; 20 of them were different breeds and sexes (test group), and the other 10 dogs with different breeds but both sexes (control group).In the test group, the blood pressure measurements were recorded on days 0, 1, 7, 14, 21 and 28th, six times a day. The control group was recorded on days 0 and 28, twice per day. Each dog?s blood pressure (SYS, DIA, MAP values) were recorded using a memodiagnostic oscillometric blood pressure device which is a certified method used in the field of veterinary medicine. The test group dogs were given a 0.5 mg/ kg single daily dose of perindopril and 1 mg/kg dose of i.m. furosemid for 3 days. Both test and control group dogs had blood drawn twice per day on days 0 and 28 for hematological and serum biochemical measurements. Samples taken for the complete blood cell count, the amount of Hgb, RBC, PLT and WBC counts, serum biochemical markers for the control of CK, LDH, AST, CK-MB and cardiac troponin-I enzyme and Na+, K+, Cl- ion values were determined.The control group of dogs did not show any presence of heart failure and hypertension during the physical examiniation.On the first day, it was determined that SYS, DIA and MAP values for of the test group dogs having heart failure and hypertension were much more higher than the same values of the dogs in the control group. On the 28th day of drug administration, all dogs in the test group had diagnostic values (SYS, DIA, MAP and blood pressure) close to the dogs in the control group and the values determined within the physiological limits.Blood table and blood serum values in dogs given perindopril did not not constitute significant changes.This study concluded the effectiveness of daily oral 0.5 mg/kg dose perindopril, an ACE inhibitor, for the treatment of dogs diagnosed with hypertension and heart failure in order to have continued levels of healthy blood pressure.
