Abstract:

In this thesis, a carbon nanotube based Myelin Basic Protein immunosensor which is able to determine Anti-Myelin Basic Protein, a auto-antibody causes Multiple Sclerosis, rapidly, cost effectively and reliably and provide clinical data in order to supports magnetic resonance imaging, was developed. For this purpose, carbon nanotubes were functionalized with polyethylene glycol and immobilized onto gelatin modified screen printed electrodes and immunosensor system were completed by immobilizing Myelin Basic Protein on this platform. Optimization and characterization of immunosensor were inspected with electrochemical impedance spectroscopy (EIS), cyclic voltammetry (CV) and scanning electron microscopy (SEM). Carbon nanotube functionalization process were characterized with Fourier-Transform Infrared Spectroscopy (FTIR) and scanning electron microscopy. Characterization investigations which were carried out with EIS, CV and SEM show that bioreceptor molecule (MBP) was successfully immobilized onto surface. Calibration curves were obtained following defining of optimum conditions. Calibration studies proved that immunosensor has a low limit of detection and wide ranges of linearity. Caibration curves were obtained and applicability of immunosensor to real samples were examined. By completion of studies, an immunosensor which is able to determine of a MS biomarker, Anti-MBP, rapidly and sensitive, was proposed.
