Abstract:

Alpha lipoic acid is fat and water soluble antioxidant that prevent damage from free radicals. Recent studies suggested that lipoic acid has anticancer potential and may inhibit cell proliferation in various types of cancer and may increase apoptosis in cancer cell lines. Many studies showed that piperlongumine, a natural alkaloid, has antitumor activity and has antiproliferative effect on tumor cells without harming healthy cells even at low doses. The purpose of this study is to investigate the effect of alpha lipoic acid and piperlongumine in lung cancer cells. In this study A549 lung cancer cells treated with various concentration of piperlongumine and alpha lipoic acid. Cell viability was determined by MTT assay. Also bax and survivin protein expression levels were determined. According to the results piperlongumine has antiproliferative effect even at low doses, whereas alpha lipoic acid has proliferative effect at low doses and antiproliferative effect at high doses. Also our results showed that alpha lipoic acid inhibits the antiproliferative effect of piperlongumine. According to western blot results bax expressions were found unaltered. Survivin, an apoptosis inhibitor protein, expressions were decreased in piperlongumine treated cells but no significant change was observed in piperlongumine+alpha lipoic acid treated cells. In the present study we found that piperlongumine has strong antiporliferative effect on lung cancer cells. Despite that alpha lipoic acid has inhibits antiproliferative effect of piperlongumine.
